Double-setting alpha-tricalcium phosphate cement provided with interconection channels in rabbits after enucleation: A potential implant for the anophthalmic socket Arq. Bras. Med. Vet. Zootec.
Gomes Junior,D.C.; Oriá,A.P.; Dórea Neto,F.A.; Santos,L.A. dos; Estrela-Lima,A.; Barbosa,V.F.; Costa Neto,J.M.; Laus,J.L.; Daleck,C.R..
The purpose of this study was to evaluate the macroscopy and microstructure of a double setting alpha-tricalcium phosphate bone cement sphere provided with interconnection channels (alpha-TCP-i), as well as the integration of the implant with the rabbits' orbital tissue, through macroscopic analysis and histopathology. The external and internal surfaces of the alpha-TCP-i were evaluated macroscopically and by electron microscopy. Twelve New Zealand rabbits received 12mm implants of alpha-TCP-i following enucleation of the left eye. The clinical assessment was undertaken daily during the first 15 days, followed by fortnightly assessment until the end of the study period. For the morphological analysis, exenteration was performed in 3 animals per...

Immunization by subcutaneous implants of polyester-polyurethane sponges coupled with antigen BJMBR
Lima,S.L.; Machado,C.B.; Pereira,M.A.C.; Cara,D.C.; Velarde,D.T.; Andrade,S.P.; Gontijo,C.M..
A new protocol is described for immunization of outbred Swiss mice. The procedure is based on subcutaneous implantation of antigen-coupled polyester-polyurethane sponges cut into disks of 10 mm in diameter vs 2 mm in thickness. Antigen coupling was performed by overnight incubation of the sponge with a solution of ovalbumin (Ova) (2 mg/ml) diluted in sodium carbonate buffer, pH 9.6. The amount of ovalbumin that was taken up by the sponge was between 71.4 to 82.5 µg. This was estimated by comparing the Ova absorbance at 280 nm in coating buffer solutions before and after incubation. To compare the efficiency of the proposed method, experimental groups immunized with the antigen in the presence of adjuvants (10 µg in Al(OH)3 or 100 µg in complete Freund's...

In vitro culture and characterization of alveolar bone osteoblasts isolated from type 2 diabetics BJMBR
Sun,Dao-Cai; Li,De-Hua; Ji,Hui-Cang; Rao,Guo-Zhou; Liang,Li-Hua; Ma,Ai-Jie; Xie,Chao; Zou,Gui-Ke; Song,Ying-Liang.
In order to understand the mechanisms of poor osseointegration following dental implants in type 2 diabetics, it is important to study the biological properties of alveolar bone osteoblasts isolated from these patients. We collected alveolar bone chips under aseptic conditions and cultured them in vitro using the tissue explants adherent method. The biological properties of these cells were characterized using the following methods: alkaline phosphatase (ALP) chemical staining for cell viability, Alizarin red staining for osteogenic characteristics, MTT test for cell proliferation, enzyme dynamics for ALP contents, radio-immunoassay for bone gla protein (BGP) concentration, and ELISA for the concentration of type I collagen (COL-I) in the supernatant....

PLGA-corosolic acid implants for potential application in ocular neovascularization diseases BJPS
Toledo,Cibele Rodrigues; Pereira,Vinícius Viana; Andrade,Gracielle Ferreira; Silva-Cunha,Armando.
Angiogenesis is the formation of new blood vessels from preexisting vasculature. Uncontrolled angiogenesis is associated with progression of several ocular pathologies, such as diabetic retinopathy and macular degeneration. Thus, the inhibition of this process consists in an interesting therapeutic target. Corosolic acid (CA) is a natural derivative of ursolic acid, found in many medicinal herbs and exhibits numerous biological properties, including the antiangiogenic activity. The present study reports the production of CA-loaded poly d,l-lactidecoglycolide acid (PLGA) devices by melt technique. HPLC-UV method was developed and validated to evaluate the uniformity and the release profile of the developed systems. The devices were also characterized by...

Resistance of oxidative stress in biofilm and planktonic cells BABT
Jakubowski,Witold; Walkowiak,Bogdan.
This work studied the susceptibility of biofilm produced by E. coli to oxidative stress, and compared the components of free radicals defences: level of glutathione, catalase and dismutase activities in planktonic and biofilm located cells. Results showed the diversity of responses to oxidative stress in bacterial cells in log or stationary phases in both planktonic and biofilm forms. The bacteria were exposed to free-radical donors (H2O2, tBOOH, menadione, SIN-1 or peroxynitrite) in a wide range of final concentrations, from 0.5 to 10mM. Different level of toxicity of individual donors, independence of cell type (planktonic forms or biofilm) and phases of growth were observed. The highest oxidative stress resistance was observed for the cells in...
